Abstract:
(EN) The purposes of the present invention are to: demonstrate, through an in vivo experiment inducing a colitis-associated cancer (CAC) initiated by azoxymethane (AOM)and promoted by dextran sodium sulfate (DSS), that an omega-3 polyunsaturated fatty acid (ω-3 PUFA) has anticancer mechanisms, such as selective cytotoxicity against colon cancer cells by means of apoptosis and cell cycle arrest, inhibition of β-catenin complex degradation, and inducement of a tumor-inhibiting 15-PGDH; and complete the useful information, for future clinical applications, that the dietary intake of 3g of the omega-3 polyunsaturated fatty acid per 60kg of weight can result in the same ω-6 : ω-3 ratio in tissue as that seen in the colon of a fat-1 mouse. The present invention provides an anticancer composition for reducing colitis-associated cancer development and treating such cancer, comprising an ω-3 polyunsaturated fatty acid as an active ingredient, wherein 3g of the ω-3 polyunsaturated fatty acid as an active ingredient is administered per 60kg per day.
(FR) Les objectifs de la présente invention sont de démontrer, grâce à une expérience in vivo induisant un cancer associé à la colite (CAC) déclenché par l'azoxyméthane (AOM) et favorisé par du sulfate de dextrane sodique (DSS), qu'un acide gras polyinsaturé de type oméga-3 (AGPI ω-3) est à l'origine de mécanismes anticancéreux, tels qu'une cytotoxicité sélective contre les cellules du cancer du côlon par apoptose et arrêt du cycle cellulaire, une inhibition de la dégradation du complexe de la β-caténine et une induction de la production de 15-PGDH inhibant les tumeurs; et de compléter les informations utiles, pour de futures applications cliniques, selon lesquelles l'apport alimentaire de 3 g d'acide gras polyinsaturé oméga-3 par 60 kg de poids peut conduire au même rapport ω-6/ω-3 dans le tissu que celui observé dans le côlon d'une souris fat-1. La présente invention concerne une composition anticancéreuse visant à inhiber le développement du cancer associé à la colite et à traiter un tel cancer, comprenant un acide gras polyinsaturé de type ω-3 en tant que principe actif, de l'acide gras polyinsaturé ω-3 étant administré en tant que principe actif à hauteur de 3 g par 60 kg et par jour.
(KO) azoxymethane(AOM)으로 개시되고, dextran sodium sulfate(DSS)의 촉진에 의해 대장염-관련 암(CAC)를 유발시킨 생체 내 실험을 통해 세포사멸과 세포주기 억제에 의한 대장암 세포에 대한 선택적 세포독성, β-카테닌 복합체의 분해 억제, 종양 억제성 15-PGDH의 유도 현상을 등의 항암 메커니즘을 오메가-3 폴리불포화지방산(ω-3 PUFA)이 가지고 있음을 밝혀내고, 또한, 장래의 임상적 활용을 위해서, 중 60kg 당 3g의 오메가-3 폴리불포화지방산을 식이 섭취하는 것이 조직에서 fat-1 생쥐의 대장에서 나타난 것과 같은 ω-6:ω-3 비율을 보일 수 있다는 유용한 정보를 완성하게 본 발명에 이르게 된 것으로, 본 발명은 ω-3 폴리불포화지방산을 활성 성분으로 포함하는 것을 특징으로 하는 대장염-관련 암 발생을 줄이고 이를 치료하는 항암 조성물을 제공하며, 특히 활성 성분으로서 상기 ω-3 폴리불포화지방산이 1일 60kg 당 3g 투여되는 것으로 특징으로 한다.
