Abstract:
(EN) Provided is a new type serum-free medium. The medium comprises: DMEM with high glucose (the content of glucose being 4.5 g/L), B27, recombinant human basic fibrolast growth factor (b-FGF), nicotinamide, N-2, vinblastine III (conophylline), non-essential amino acid (NEAA), heparin, epidermal growth factor (EGF), hepatocyte growth factor (HGF), a serum replacement (SR), an insulin-transferrin-selenium complex (ITS), and pentagastrin. Inducing differentiation of mesenchymal stem cells into insulin-secretion-like cells can be achieved in six days in one step using the medium.
(FR) L'invention concerne un nouveau milieu exempt de sérum. Le milieu comprend : du DMEM à teneur élevée en glucose (la teneur en glucose étant de 4,5 g/l), du B27, le facteur de croissance du fibroblaste basique humain recombinant (b-FGF), du nicotinamide, du N-2, de la vinblastine III (conophylline), un acide aminé non essentiel (NEAA), de l'héparine, le facteur de croissance épidermique (EGF), le facteur de croissance des hépatocytes (HGF), un remplacement sérique (SR), un complexe insuline-transferrine-sélénium (ITS) et une pentagastrine. L'induction de la différenciation de cellules souches mésenchymateuses en cellules de type à sécrétion d'insuline peut être obtenue en six jours en une étape à l'aide du milieu.
(ZH) 一种新型无血清培养基。该培养基包含:高糖的DMEM(葡萄糖含量4.5g/L)、B27、重组人碱性成纤维生长因子(b-FGF)、尼古酰胺、N-2、长春花碱Ⅲ(Conophylline)、非必需氨基酸(NEAA)、肝素(Heparin)、表皮细胞生长因子(EGF)、肝细胞生长因子(HGF)、血清替代物(SR)、胰岛素-转铁蛋白-硒复合物(ITS)、五肽胃泌素(Pentagastrin)。利用该培养基可实现在6天内一步法将间充质干细胞诱导分化为向胰岛素分泌样细胞。
