(EN)Provided herein are compositions and methods for generation of naive human pluripotent stem cells. The method comprises incubation of iPSCs under 5% O2 in a medium comprising 5% glucose, an MEK inhibitor, a GSK3β inhibitor, human leukemia inhibitory factor (LIF), human insulin and Torin 1. The method does not need any other inhibitors or transgene expression. The naive human pluripotent cells can be used to generate a large amount of mature human cells from all three germ layers in host non-human animals.
(FR)La présente invention concerne des compositions et des procédés de production de cellules souches pluripotentes humaines naïves. Le procédé comprend l’incubation d’iPSC sous O2 à 5 % dans un milieu comprenant du glucose à 5 %, un inhibiteur MEK, un inhibiteur GSK3P, le facteur d’inhibition de la leucémie humaine (LIF), de l’insuline humaine et de la torine (1). Le procédé ne nécessite aucuns autres inhibiteurs ni expression transgénique. Les cellules pluripotentes humaines naïves peuvent être utilisées pour générer une grande quantité de cellules humaines matures à partir de l'ensemble des trois couches germinales chez des animaux hôtes non humains.
