Abstract:
(EN) Disclosed is a quantitative cluster analysis method of target proteins by using next-generation sequencing. The method of the present invention comprises the steps of: (a) treating a sample to be analyzed with a library of aptamers which directs toward a group of target proteins present in the sample so as to form complexes between target proteins and aptamers binding specifically thereto, with the consequent formation of a complex cluster of the target proteins and aptamers; (b) separating the complex cluster from unbound aptamers; and (c) analyzing a sequence of each of the aptamers in the complex cluster by a next-generation sequencing analysis technique so as to quantify each of the aptamers in the complex cluster and thereby to quantify each of target proteins in the complex cluster. The method of the present invention can be very useful in collectively quantifying proteins in an analysis sample.
(FR) La présente invention concerne un procédé d'analyse quantitative de clusters de protéines cibles à l'aide d'un séquençage de nouvelle génération. Le procédé selon la présente invention comprend les étapes suivantes : (a) traiter l'échantillon à analyser avec une banque d'aptamères qui cible un groupe de protéines cibles présentes dans l'échantillon pour former des complexes entre les protéines cibles et les aptamères se liant spécifiquement à celles-ci, avec formation ultérieure d'un cluster complexe de protéines cibles et d'aptamères; (b) séparer le cluster complexe des aptamères non liés; et (c) analyser une séquence de chacun des aptamères dans le cluster complexe par une technique d'analyse de séquençage de nouvelle génération pour quantifier chacun des aptamères dans le cluster complexe, chacune des protéines cibles dans le cluster complexe étant analysée de manière quantitative. Le procédé selon la présente invention peut être très utile dans la quantification collective de protéines dans un échantillon d'analyse.
(KO) 본 발명은 차세대서열결정법을 이용한 표적 단백질의 집단적 정량 방법을 개시한다. 본 발명의 방법은 (a) 분석 대상 시료에, 그 시료에 존재하는 표적 단백질 집단에 압타머 라이브러리를 처리하여, 각 표적 단백질과 이에 특이적으로 결합하는 각 압타머 사이의 복합체를 형성시킴으로써, 표적 단백질과 압타머 사이의 복합체 집단을 형성시키는 단계와, (b) 상기 복합체 집단을 비결합 압타머로부터 분리하는 단계와, (c) 상기 복합체 집단의 각 압타머의 서열을 차세대서열분석 기술로 분석하여 그 복합체 집단의 각 압타머를 정량함으로써 상기 복합체 집단의 각 표적 단백질을 정량하는 단계를 포함한다. 본 발명의 방법은 분석 시료 중의 단백질들을 집단적으로 정량하는데 매우 유용하게 사용될 수 있다.
