(EN)The screening system disclosed in the present specification is provided with a potential-dependent Na ion channel that extends the duration of the action potential associated with depolarization, and a K ion channel that deepens the resting membrane potential in the negative direction, said screening system furthermore including cells provided with ion channels that contribute to deepening the resting membrane potential in the negative direction and/or shortening the duration of the action potential as target ion channels. By such cells, the action of a test compound on the target ion channel can be easily evaluated by providing an inhibitor for the K ion channel to control the probability of cell death.
(FR)Cette invention concerne un système de criblage pourvu d'un canal ionique Na potentiel-dépendant qui prolonge la durée du potentiel d'action associé à la dépolarisation, et d'un canal ionique K qui approfondit le potentiel de repos de la membrane dans le sens négatif, ledit système de criblage comprenant en outre des cellules pourvues de canaux ioniques qui contribuent à approfondir le potentiel de repos de la membrane dans le sens négatif et/ou à raccourcir la durée du potentiel d'action à titre de canaux ioniques cibles. Ces cellules permettent d'évaluer facilement l'action d'un composé d'essai sur le canal ionique cible par utilisation d'un inhibiteur de canal ionique K permettant de contrôler la probabilité de mort cellulaire.
