Abstract:
(EN) A glycan having a branched structure is labeled using a labeling agent, such as 2-aminobenzoic acid, that has one site that is easily negatively charged. At this time, the reduction that is normally carried out in labeling through reductive amination is not carried out. A sample for mass spectrometry including the labeled glycan obtained in this way is prepared, and tandem mass spectrometry is carried out on the sample in a negative ion mode. The peaks of, for example, E ions and D ions reflecting the branch structure appear clearly in the tandem mass spectrum obtained in this way. As a result, structural analysis of the entirety of a glycan including a branched structure can be carried out easily.
(FR) Un glycane à structure ramifiée est marqué à l'aide d'un agent de marquage, tel que l'acide 2-aminobenzoïque, qui possède un site facilement chargé négativement. A ce moment, la réduction qui est normalement effectuée lors du marquage par amination réductrice n'est pas effectuée. Un échantillon pour spectrométrie de masse comprenant le glycane marqué ainsi obtenu est préparé, et une spectrométrie de masse en tandem est réalisée sur l'échantillon en mode ions négatifs. Les pics correspondant, par exemple, aux ions E et aux ions D reflétant la structure ramifiée apparaissent clairement dans le spectre de masse en tandem ainsi obtenu. Par conséquent, une analyse structurale de la totalité d'un glycane comprenant une structure ramifiée peut être réalisée facilement.
(JA) 分枝構造を有する糖鎖を、負に帯電し易い部位を一つ有する、2-アミノ安息香酸などのラベル化剤を用いてラベル化する。この際に、還元アミノ化によるラベル化では通常実施される還元を行わない。こうして得られた糖鎖のラベル化体を含む質量分析用サンプルを調製し、該サンプルについて負イオンモードでMS/MS分析を実行する。これにより得られるMS/MSスペクトルには、分枝構造を反映したEイオン、Dイオンなどのピークが明瞭に現れる。これにより、分枝構造を含む糖鎖全体の構造解析を容易に行うことができる。
