Abstract:
(EN) The present invention relates to A multicomponent system wherein a first component is an engineered antigen-presenting cell (eAPC) designated component A and a second component is a genetic donor vector, designated component C, for delivery of one or more ORFs encoding an analyte antigen-presenting complex (aAPX) and/or an analyte antigenic molecule (aAM), wherein component A a. Lacks endogenous surface expression of at least one family of aAPX and/or aAM and b. Contains at least two genomic receiver sites, designated component B and component D, each for integration of at least one ORF encoding at least one aAPX and/or aAM, and component C is matched to a component B, and wherein component C is designed to deliver c. A single ORF encoding at least one aAPX and/or aAM or d. Two or more ORF encoding at least one aAPX and/or aAM, wherein the genomic receiver sites B and D are synthetic constructs designed for re- combinase mediated exchange (RMCE).
(FR) La présente invention concerne un système à composants multiples dans lequel un premier composant est une cellule présentatrice d'antigène génétiquement modifiée (eAPC) formant le composant A et un second composant est un vecteur donneur génétique, formant le composant C, pour administration d'un ou plusieurs ORF codant pour un complexe de présentation d'antigène (aAPX) à analyser et/ou une molécule antigénique (aAM) à analyser, dans lequel le composant A : a. ne présente pas d'expression de surface endogène d'au moins une famille d'aAPX et/ou d'aAM et b. contient au moins deux sites récepteurs génomiques, formant les composants B et D, chacun en vue de l'intégration d'au moins un ORF codant pour au moins un aAPX et/ou une aAM, le composant C étant mis en correspondance avec un composant B, et le composant C étant conçu pour délivrer c. un ORF unique codant pour au moins un aAPX et/ou une aAM ou d. au moins deux ORF codant pour au moins un aAPX et/ou une aAM, les sites récepteurs génomiques B et D étant des produits de synthèse conçus pour un échange à médiation par la recombinase (RMCE).
