Abstract:
(EN) This disclosure generally relates to stable water isotope labeling followed by detection via MRI (swiMRI), including deuterium MRI (dMRI) and 17O MRI, for visualizing rapidly dividing immune cells within target and/or lymphoid organ/s and/or tissues affected by chronic graft-versus-host disease (cGVHD). Using deuterated water labeling, followed by dMRI, a distinction in deuterium signal was detected in a target organ (e.g. liver) of the cGVHD-affected mice compared to unaffected mice, i.e. syngeneic HSCT recipient mice, where the host and donor are matched, and normal (unmanipulated) mice.
(FR) La présente invention concerne d'une manière générale le marquage stable d'isotopes d'eau suivi d'une détection par IRM (IRM pondérée par susceptibilité magnétique, SWI), notamment l'IRM du deutérium (IRMd)) et IRM de 17O, pour visualiser des cellules immunitaires se divisant rapidement dans des organes et/ou des tissus cibles et/ou lymphoïdes affectés par une maladie du greffon contre l'hôte chronique (GVHc). A l'aide d'un marquage par de l'eau deutérée, suivi par une IRMd, une différence dans un signal de deutérium a été détectée dans un organe cible (par exemple, le foie) de souris atteintes de GVHc par comparaison avec des souris non affectées, à savoir des souris receveuse HSCT syngéniques, l'hôte et le donneur correspondant, et des souris normales (non manipulées).
