Abstract:
(EN) Nonnaturally occurring host cells altered to increase their ability to transfer genetic molecules into the host cells as compared to an unaltered host cell are provided. Also provided are methods for identifying endogenous loci of a host cell which inhibit transformation efficiency and/or electroporation of genetic molecules into the cell as well as methods for producing nonnaturally occurring host cells with enhanced transformation efficiency and/or the modified ability to allow for genomic integration of an exogenous DNA sequence via electroporation. Methods for producing biochemicals and products produced with the nonnaturally occurring host cells are also provided. In particular, a host Cupriavidus necator cell is provided in which a chromosomal gene encoding a restriction endonuclease has been disrupted, leading to an increase in transformation efficiency.
(FR) Des cellules hôtes ne se produisant naturellement sont modifiées pour augmenter leur capacité à transférer des molécules génétiques dans les cellules hôtes par comparaison avec une cellule hôte non modifiée. L'invention concerne également des procédés d'identification de loci endogènes d'une cellule hôte qui inhibent l'efficacité de transformation et/ou l'électroporation de molécules génétiques dans la cellule, ainsi que des procédés de production de cellules hôtes ne se produisant pas naturellement avec une efficacité de transformation améliorée et/ou la capacité modifiée de permettre l'intégration génomique d'une séquence d'ADN exogène par électroporation. L'invention concerne également des procédés de production de produits biochimiques et de produits produits avec les cellules hôtes ne se produisant pas naturellement. En particulier, l'invention concerne une cellule de Cupriavidus necator hôte dans laquelle un gène chromosomique codant pour une endonucléase de restriction a été interrompu, conduisant à une augmentation de l'efficacité de transformation.
