Abstract:
(EN) The present invention relates to a method for modifying the resistance profile of a spinach plant to Peronospora farinosa f. sp. spinaciae, comprising introducing a WOLF allele or a resistance-conferring part thereof into the genome of said spinach plant, or modifying an endogenous WOLF allele in the genome of said spinach plant. A WOLF allele encodes a CC-NBS- LRR protein that comprises in its amino acid sequence: the motif "MAEIGYSVC" at its N- terminus, and the motif "KWMCLR" for alpha-type WOLF proteins or "HVGCVVDR" for beta- type WOLF proteins. The invention relates to the WOLF alleles referred to in Table 3. The invention further provides a method for selecting a spinach plant comprising a novel WOLF gene that confers resistance to Peronospora farinosa f. sp. spinaciae in a spinach plant and a method for identifying a WOLF allele that confers resistance to one or more pathogenic races of Peronospora farinosa f. sp. spinaciae in a spinach plant and to primers for use in these methods.
(FR) L'invention concerne un procédé destiné à modifier le profil de résistance d'une plante d'épinard à Peronospora farinosa f. sp. spinaciae, consistant à introduire un allèle WOLF ou une partie de celui-ci conférant une résistance dans le génome de la plante d'épinard, ou à modifier un allèle WOLF endogène dans le génome de ladite plante. Un allèle WOLF code une protéine CC-NBS-LRR qui comprend dans sa séquence d'acides aminés : le motif "MAEIGYSVC" à son extrémité N-terminale et le motif "KWMCLR" pour les protéines WOLF de type alpha ou "HVGCVVDR" pour les protéines WOLF de type bêta. L'invention concerne les allèles WOLF mentionnés dans le tableau 3. L'invention concerne également un procédé de sélection d'une plante d'épinard comprenant un nouveau gène WOLF conférant une résistance à Peronospora farinosa f. sp. spinaciae dans une plante d'épinard et un procédé d'identification d'un allèle WOLF confèrant une résistance à au moins une race pathogène de Peronospora farinosa f. sp. spinaciae dans une plante d'épinard, ainsi que des amorces destinées à être utilisées dans ces procédés.
