Abstract:
(EN) The present invention relates to methods for assessing binding agent specificity, in particular antibody specificity. The present invention thus provides a method of analysing a mixture of polypeptides comprising the steps of: (i) separating the polypeptides in the mixture into a plurality of fractions; (ii) contacting a first aliquot of two or more of the fractions with a plurality of different binding agents attached to one or more solid supports and detecting the binding of the polypeptides to the binding agents in each fraction; (iii) assessing the amino acid composition of the polypeptides in a second aliquot of said fractions by mass spectrometry; and (iv) correlating the binding results detected in step (ii) and the mass spectrometry results from step (iii) to assess the specificity of the binding agents for a polypeptide of interest.
(FR) La présente invention concerne des procédés pour l'évaluation de la spécificité d'un agent de liaison, en particulier de la spécificité d'un anticorps. La présente invention concerne ainsi un procédé d'analyse d'un mélange de polypeptides comprenant les étapes consistant : (i) à séparer les polypeptides se trouvant dans le mélange en une pluralité de fractions ; (ii) à mettre en contact une première aliquote d'au moins deux des fractions avec une pluralité d'agents de liaison différents fixés à un ou plusieurs supports solides et à détecter la liaison des polypeptides aux agents de liaison dans chaque fraction ; (iii) à évaluer la composition en acides aminés des polypeptides dans une seconde aliquote desdites fractions par spectrométrie de masse ; et (iv) à mettre en corrélation les résultats de liaison détectés à l'étape (ii) et les résultats de la spectrométrie de masse de l'étape (iii) pour évaluer la spécificité des agents de liaison pour un polypeptide d'intérêt.
