(EN) A method for using a single-stranded ribonucleic acid to convert penicillium amagasakiense spores and manufacture a protein, said method comprising the three steps of penicillium amagasakiense culturing and spore collection, spore preprocessing, and spore shocking via an HDEN method. The method bypasses the step of fungal spore germination, HDEN electroconversion technology is used, the in vitro single-stranded coding protein RNA is delivered through cell walls and cell membranes, and the protein is expressed within the dormant penicillium amagasakiense spores.
(FR) La présente invention concerne un procédé d’utilisation d’un acide ribonucléique simple brin destiné à convertir des spores de Penicillium amagasakiense et la fabrication d’une protéine, ledit procédé comprenant les trois étapes de culture de Penicillium amagasakiense et de collecte de spores, le prétraitement des spores, et le choc des spores par l’intermédiaire d’un procédé HDEN. Le procédé contourne l’étape de germination des spores de champignon, la technologie d’électroconversion HDEN est utilisée, l’ARN protéique codant simple brin in vitro est libéré à travers les parois cellulaires et les membranes cellulaires, et la protéine est exprimée à l’intérieur des spores dormants de Penicillium amagasakiense.
