Abstract
(EN) Polypeptides that fold into biologies are stabilized by diselenide bonds between selenocysteine amino acids. Methods to produce such polypeptides in genomically recoded organisms (GRO) can be scaled up for industrial production. Since diselenides have the same geometric bond angles and torsions as disulfides, as well as very similar bond lengths, they can be substituted into polypeptides without disrupting the three dimensional structure of the polypeptides. Diselenides render the polypeptides resistant to reduction when they are exposed to blood serum or to reducing components of blood serum or to reducing components components within cells.
(FR) L'invention concerne des polypeptides se repliant en biologiques et stabilisés par des liaisons diséléniure entre les acides aminés de sélénocystéine. L'invention concerne également des procédés de production de ces polypeptides dans des organismes génétiquement recodés (OGR), lesdits procédés étant adaptés pour une production industrielle. Étant donné que les diséléniures ont les mêmes angles de liaison géométriques et les mêmes torsions que les disulfures, ainsi que des longueurs de liaison très similaires, ils peuvent servir d'éléments de substitution dans des polypeptides sans perturber la structure tridimensionnelle des polypeptides. Les diséléniures rendent les polypeptides résistants à la réduction lorsqu'ils sont exposés au sérum sanguin ou à des composants réducteurs du sérum sanguin, ou à des composants réducteurs au sein des cellules.
