Abstract:
(EN) The invention describes composition and methods of use for novel hairpin blocked-cleavable primers. In one embodiment unblocking occurs through action of RNase H2. The method improves the specificity of PCR and reduces primer dimer events, enabling higher level multiplex reactions. Additionally, the invention protects RNA-containing primers from attack by single-strand RNases.
(FR) L'invention concerne une composition et des procédés d'utilisation de nouvelles amorces clivables bloquées en épingle à cheveux. Dans un mode de réalisation, le déblocage se produit via l'action de l'ARNase H2. Le procédé améliore la spécificité de la PCR et réduit les événements de dimérisation d'amorce, ce qui permet des réactions de multiplexage de niveau supérieur. De plus, l'invention protège les amorces contenant de l'ARN contre une attaque par des ARNases à simple brin.
