Title: METHOD FOR PRODUCING MUTANT ENZYME, AND MUTANT ALCOHOL ACYLTRANSFERASE
Abstract:
Provided is a method for producing an enzyme having higher activity per recombinant than does the standard form, the method including the following steps, as an efficient method for causing a target protein to be expressed in a transformant as an active soluble recombinant protein. (1) A step for producing a recombinant that expresses a mutant having an amino acid sequence in which one or more cysteines have been substituted by other amino acid residues in the amino acid sequence of the standard form; (2) a step for selecting a plurality of mutants that show activity per recombinant of 50% or higher in comparison to 100% activity of the standard form; and (3) a step for inducing expression of a mutant in which corresponding amino acid residues are substituted at two or more sites among the sites at which amino acid residues were substituted in each of the mutants selected in step (2).
