Abstract:
(EN) Provided are a glycoside screening method and a glycoside quantification method, each comprising: preparing both of an analysis sample that is treated with an enzyme capable of recognizing a sugar structure of a glycoside to hydrolyze a glycoside bond and an analysis sample that is not treated with the enzyme, from a preparation prepared from a biological sample and containing a glycoside; comparing the results of the analysis of these analysis samples by LC-MS/MS or CE-MS/MS; and determining that a peak which disappears or of which the intensity (height or area) decreases as the result of the treatment with the enzyme comes from the glycoside.
(FR) La présente invention concerne un procédé de criblage de glycoside et un procédé de quantification de glycoside, comprenant chacun : la préparation à la fois d’un échantillon d’analyse qui est traité avec une enzyme capable de reconnaître une structure de sucre d’un glycoside pour hydrolyser une liaison glycosidique et un échantillon d’analyse qui n’est pas traité avec l’enzyme, à partir d’une préparation préparée à partir d’un échantillon biologique et contenant un glycoside ; la comparaison des résultats de l’analyse de ces échantillons d’analyse par LC-MS/MS ou CE-MS/MS ; et la détermination qu’un pic qui disparaît ou dont l’intensité (hauteur ou aire) diminue en conséquence du traitement avec l’enzyme provient du glycoside.
(JA) 生物試料より調製された配糖体を含む調製物から、配糖体の糖構造を認識してグリコシド結合を加水分解する酵素で処理した分析用試料と酵素で処理しない分析用試料を準備して、それぞれをLC-MS/MSまたはCE-MS/MSにより分析した結果を比較して、酵素処理により消失または強度(高さまたは面積)が減少しているピークを配糖体由来であると決定する、配糖体のスクリーニング方法および定量方法を提供する。
