Abstract:
(EN) The present invention includes a novel method and system for identification of microorganisms in samples that proteins and other biological material from non-microorganism sources (e.g., proteins of mammalian origin) that can interfere with identification of the microorganisms. The methods and systems described herein include use of a single-use chromatography medium to purify intact proteins prior to mass spectrometry analysis. The chromatography medium and the methods described herein can rapidly and efficiently remove of a substantial portion of interfering biological material (e.g., mammalian proteins) from a crude cell lysate while preserving high signal strength and removing enough of the interfering protein(s) to allow for identification of the microorganism(s) by mass spectrometry analysis.
(FR) La présente invention concerne un nouveau procédé et un nouveau système d’identification de micro-organismes dans des échantillons qui contiennent des protéines et d’autres matériaux biologiques provenant de sources non micro-organisme (par exemple, des protéines d’origine mammalienne) qui peuvent interférer avec l’identification des micro-organismes. Les procédés et les systèmes de l’invention comprennent l’utilisation d’une phase de chromatographie à usage unique pour purifier des protéines intactes avant une analyse par spectrométrie de masse. La phase de chromatographie et les procédés de l’invention peuvent éliminer rapidement et efficacement une partie substantielle de matériel biologique interférant (par exemple, des protéines de mammifère) d’un lysat cellulaire brut tout en conservant une intensité de signal élevée et en éliminant suffisamment de(s) protéine(s) interférente(s) pour permettre l’identification du ou des micro-organisme(s) par analyse par spectrométrie de masse.
