(KO) 유도만능줄기세포의 분화를 유도하여 각막 상피 세포를 배양하는 방법 및 시스템
Abstract:
(EN) The present invention provides a method for culturing a corneal epithelial cell by inducing the differentiation of an induced pluripotent stem cell, the method comprising the steps of: preparing a basal medium comprising DMEM F12, L-ascorbic acid, sodium selenite and sodium chloride; adding, to the basal medium, an additive for culturing an induced pluripotent stem cell; producing a feeder-free culture medium by coating the basal medium with a vitronectin recombinant human protein; adding, to the feeder-free culture medium, an induced pluripotent stem cell and culturing same; adding, to the feeder-free culture medium, an additive for creating an environment for stem cell growth; consecutively adding BMP4 and KGF to the feeder-free culture medium in order to induce the differentiation of the induced pluripotent stem cell into a corneal epithelial cell; and culturing, in a PI culture fluid, the cell differentiated from the induced pluripotent stem cell.
(FR) L'invention concerne un procédé de culture d'une cellule épithéliale cornéenne par induction de la différenciation d'une cellule souche pluripotente induite, ce procédé comprenant les étapes consistant : à préparer un milieu de base comprenant du DMEM F12, de l'acide L-ascorbique, du sélénite de sodium et du chlorure de sodium ; à ajouter au milieu de base un additif pour la culture d'une cellule souche pluripotente induite ; à produire un milieu de culture sans cellules nourricières par revêtement du milieu de base au moyen d'une protéine humaine recombinée de vitronectine ; à ajouter au milieu de culture sans cellules nourricières une cellule souche pluripotente induite et à la cultiver ; à ajouter au milieu de culture sans cellules nourricières un additif destiné à créer un environnement favorable à la croissance de la cellule souche ; à ajouter successivement BMP4 et KGF au milieu de culture sans cellules nourricières afin d'induire la différenciation de la cellule souche pluripotente induite en une cellule épithéliale cornéenne ; et à cultiver, dans un fluide de culture PI, la cellule différenciée de la cellule souche pluripotente induite.
(KO) 본 발명은 유도만능줄기세포(Induced Pluripotent Stem Cell)의 분화를 유도하여 각막 상피 세포를 배양하는 방법으로서, DMEM F12, L-아스코르브산, 아셀렌산 나트륨(Sodium selenite), 및 염화 나트륨을 포함하는 기본 배지를 준비하는 과정; 상기 기본배지에 유도만능줄기세포 배양용 첨가제를 첨가하는 과정; 상기 기본배지에 비트로넥틴 재조합 인간 단백질을 코팅하여 피더 비포함 배지(feeder free culture medium)를 제조하는 과정; 상기 피더 비포함 배지에 유도만능줄기세포를 첨가하여 배양하는 과정; 상기 피더 비포함 배지에 줄기세포 성장 환경 조성용 첨가제를 첨가하는 과정; 상기 유도만능줄기세포를 각막 상피 세포로 분화하도록 유도하기 위해, 피더 비포함 배지에 BMP4 및 KGF를 순차적으로 첨가하는 과정; 및 상기 유도만능줄기세포로부터 분화된 세포를 PI 배양액에서 배양하는 과정;을 포함하는 방법을 제공한다.
