The present invention relates to the treatment of glioma. Considering the potential of anti-GIC therapies to advance GBM treatment, the inventors employed a chemical screen of currently FDA- and EM A- approved small molecules in order to identify a selective inhibitor of GIC (the four genetic subtypes of GBM) expansion with known bio-safety in humans. The cytotoxic effect of the compounds was initially evaluated in two patient-derived GIC growing as spheres and identified β-escin, a mixture of triterpenoid saponins isolated from the seeds of horse chestnuts. They demonstrated a specific effect of β-escin on induction of apoptosis and modulation of sternness properties of nine individual GIC, indicating the potential of β-escin as a selective and potent inhibitor of GIC. Thus the invention relate to an escin family compound (and particularly β-escin) for use in the treatment of glioma.
