Expressing guide nucleic acids (e.g., gRNA) from the same oligonucleotide that contains donor sequence permits the high efficiency, simultaneous transformation of a population of cells with both substrates. Using oligonucleotide chip array technology, one can construct thousands of oligonucleotides with customized gRNA and donor sequence in a cost effective manner. In combination, one can efficiently modify endogenous and exogenous genes.

Selon l'invention, l'expression d'acides nucléiques guides (par exemple ARNg) à partir du même oligonucléotide contenant une séquence donneur permet la transformation simultanée et à haut rendement d'une population de cellules à l'aide des deux substrats. Au moyen de la technologie des matrices de puces oligonucléotidiques, on peut construire de manière rentable des milliers d'oligonucléotides avec une séquence d'ARNg et donneur personnalisée. En combinaison, on peut modifier de façon efficace des gènes endogènes et exogènes.
