Abstract:

Provided is a method for amplifying cyclic DNA, particularly long-chain cyclic DNA, simply and exponentially in a cell-free system. More specifically, a cyclic DNA amplification method is provided, which comprises allowing to react a reaction mixture produced by mixing cyclic DNA having an origin sequence of chromosomal replication (an origin of chromosome (oriC)) with a reaction solution, wherein the reaction solution contains enzymes (1) to (3) as mentioned below, a buffer solution, NTP, dNTP, a magnesium ion source and an alkali metal ion source: (1) a first group enzyme which can catalyze the replication of cyclic DNA; (2) a second group enzyme which can catalyze an Okazaki fragment connection reaction to synthesize two sister cyclic DNA molecules capable of forming a catenane; and (3) a third group enzyme which can catalyze a reaction of separating two sister cyclic DNA molecules from each other.

L'invention concerne un procédé d'amplification d'ADN cyclique, en particulier d'ADN cyclique à chaîne longue, de manière simple et exponentielle dans un système acellulaire. Plus spécifiquement, l'invention porte sur un procédé d'amplification d'ADN cyclique, qui consiste à permettre la réaction d'un mélange réactionnel produit par mélange d'ADN cyclique ayant une séquence d'origine de réplication chromosomique (une origine du chromosome (oriC)) avec une solution de réaction, la solution de réaction contenant des enzymes (1) à (3) comme mentionné ci-dessous, une solution tampon, NTP, dNTP, une source d'ions magnésium et une source d'ions de métal alcalin : (1) une enzyme du premier groupe qui peut catalyser la réplication de l'ADN cyclique ; (2) une enzyme du second groupe qui peut catalyser une réaction de liaison de fragments d'Okazaki pour synthétiser deux molécules d'ADN cyclique sœurs capables de former une caténane ; et (3) une enzyme du troisième groupe qui peut catalyser une réaction de séparation entre deux molécules d'ADN cyclique sœurs.
