The present invention relates to a method for evaluating,
in vivo
, the activity of an RNA-guided nuclease in a high-throughput manner and, more particularly, to a method for evaluating the activity of an RNA-guided nuclease by using indel frequencies of a cell library comprising separate oligonucleotides including a guide RNA-encoding base sequence and a target base sequence. According to the present invention, an RNA-guided nuclease characteristic analysis method using a library of guide RNA-target sequence pairs can evaluate, in vivo, the activity of the RNA-guided nuclease in a high-throughput manner, thereby being very useful in all fields in which an RNA-guided nuclease is applied.

La présente invention concerne un procédé d'évaluation,
in vivo
, de l'activité d'une nucléase guidée par ARN de manière très efficace et, plus particulièrement, un procédé d'évaluation de l'activité d'une nucléase guidée par ARN en utilisant les fréquences d’insertion/de délétion (indel) d'une bibliothèque de cellules comprenant des oligonucléotides séparés comprenant une séquence de base de codage d'ARN de guidage et une séquence de base cible. Selon la présente invention, un procédé d'analyse de caractéristique de nucléase guidée par ARN utilisant une bibliothèque de paires de séquences cibles ARN guidée peut évaluer, in vivo, l'activité de la nucléase guidée par ARN de manière très efficace, étant ainsi très utile dans tous les domaines dans lesquels une nucléase guidée par ARN est appliquée.
