Abstract:
(EN) Disclosed are: a culture method for pancreatic precursor cells derived from pluripotent stem cells, said culture method comprising step (A) for three-dimensionally culturing pancreatic precursor cells derived from pluripotent stem cells in a medium that contains a factor belonging to the epidermal growth factor (EGF) family and/or a factor belonging to the fibroblast growth factor (FGF) family, and (2) a Wnt agonist; a production method for islet cells from pancreatic precursor cells derived from pluripotent stem cells, said production method comprising step (E) for inducing the differentiation of the pancreatic precursor cells cultured by the aforesaid method into islet cells; and a cryopreservation method for pancreatic precursor cells derived from pluripotent stem cells, said cryopreservation method comprising step (F) for freezing the pancreatic precursor cells cultured by the aforesaid method.
(FR) La présente invention concerne : un procédé de culture de cellules précurseurs pancréatiques dérivées de cellules souches pluripotentes, ledit procédé de culture comprenant l’étape (A) de culture tridimensionnelle de cellules précurseurs pancréatiques dérivées de cellules souches pluripotentes dans un milieu qui comporte un facteur appartenant à la famille des facteurs de croissance épidermique (EGF) et/ou un facteur appartenant à la famille des facteurs de croissance des fibroblastes (FGF), et (2) un antagoniste Wnt ; un procédé de production destiné aux cellules d’îlot provenant de cellules précurseurs pancréatiques dérivées de cellules souches pluripotentes, ledit procédé de production comprenant l’étape (E) d’induction de la différenciation des cellules précurseurs pancréatiques cultivées par le procédé susmentionné en cellules d’îlot ; et un procédé de cryoconservation destiné aux cellules précurseurs pancréatiques dérivées des cellules souches pluripotentes, ledit procédé de cryoconservation comprenant l’étape (F) de congélation des cellules précurseurs pancréatiques cultivées selon le procédé susmentionné.
