Abstract:
(EN) The subject invention pertains to materials and methods of determining accumulated mutations and the rate of mutations in a target genomic sequence which is a part of a short interspersed element (SINE). The assay utilizes a combination of a target sequence clamp and digital PCR (dPCR). The target sequence clamp prevents PCR amplification of the wild- type target sequence and permits PCR amplification of only the mutated target sequence. The dPCR provides the number of mutated target sequences per genome, which can be used to determine the rate of mutations in the target sequence, the accumulated mutations in the genome and/or the rate of mutations in the genome. The accumulated mutations and the rate of mutations in the target sequence and/or the genome can be used to determine the genomic age and/or the risk of cancer of a subject. A kit for performing the assay is also provided.
(FR) La présente invention concerne des matériels et des méthodes de détermination de mutations accumulées et du taux de mutations dans une séquence génomique cible qui fait partie d'un élément intercalé court (SINE). Le dosage utilise une combinaison d'une pince de séquence cible et d'une PCR numérique (dPCR). Ladite pince de séquence cible empêche l'amplification par PCR de la séquence cible de type sauvage et ne permet l'amplification par PCR que de la séquence cible ayant subi une mutation. La dPCR fournit le nombre de séquences cibles ayant subi une mutation par génome, qui peut être utilisé pour déterminer le taux de mutations dans la séquence cible, les mutations accumulées dans le génome et/ou le taux de mutations dans le génome. Les mutations accumulées et le taux de mutations dans la séquence cible et/ou le génome peuvent être utilisés pour déterminer l'âge génomique et/ou le risque de cancer chez un sujet. L'invention concerne également un kit permettant de réaliser le dosage.
