(EN)The present invention relates to heterogeneous niche activity in mesenchymal stromal cell-based cell therapy. It was discovered in the present invention that a difference in the niche activities of MSCs can be created during ex-vivo expansion of MSCs to cause a variation in the outcomes of hematopoietic recoveries. Particularly, the difference is caused by the functional state of MSCs derived by distinct upstream signaling pathways, rather than by clonal heterogeneity, and the functional state can be inferred through the CFU-F of MSC. Therefore, the present invention is expected to contribute to solving a variation in therapeutic effects which is pointed out as a problem of conventional mesenchymal stromal cell-based cell therapy.
(FR)La présente invention se rapporte à une activité de niche hétérogène en thérapie cellulaire à base de cellules stromales mésenchymateuses (MSC). Il a été découvert dans la présente invention qu'une différence des activités de niche de MSC peut être créée pendant l'expansion ex vivo de MSC pour provoquer une variation des résultats de récupérations hématopoïétiques. En particulier, la différence est provoquée par l'état fonctionnel de MSC dérivées par des voies de signalisation amont distinctes, plutôt que par hétérogénéité clonale, et l'état fonctionnel peut être déduit des CFU-F de MSC. Par conséquent, on s'attend à ce que la présente invention contribue à résoudre une variation des effets thérapeutiques qui est signalée en tant que problème de la thérapie cellulaire à base de cellules stromales mésenchymateuses classiques.
(KO)본 발명은 중간엽 기질세포-기반 세포 치료에서의 이질적인 니치 활성에 관한 것이다. 본 발명에서는 MSC의 니치 활성 차이는 ex-vivo 팽창 단계에서 이루어지며, 조혈 회복에서의 다양한 결과로 이어질 수 있음을 보여주었다. 특히, 이러한 차이는 클론의 이질성으로부터 기인하기 보다는, 고유의 상류 신호 경로에 의해 유도되는 MSC의 기능적 상태로부터 기인하며, 이러한 기능적 상태는 MSC의 CFU-F를 통해 유추할 수 있었다. 따라서, 본 발명은 종래의 중간엽 기질세포-기반 세포 치료의 문제점으로 지적되고 있는 치료 효과의 가변성을 해결함에 기여할 것으로 기대된다.
