(JA) 人工多能性幹細胞の腸管上皮細胞への分化誘導
Abstract:
(EN) This invention addresses the problem of providing: a novel method that allows, in a simple manner, preparation of cells that exhibit a function similar to that of intestinal epithelial cells of a living body; and an application therefor. Induced pluripotent stem cells are induced to differentiate into intestinal epithelial cells by: (1) a step for differentiating the induced pluripotent stem cells into endoderm-like cells; (2) a step for differentiating the endoderm-like cells obtained in step (1) into intestinal stem cell-like cells; and (3) a step for differentiating the intestinal stem cell-like cells obtained in step (2) into intestinal epithelial cell-like cells, the step including culturing in the presence of an MEK1 inhibitor, a DNA methylation inhibitor, a TGFβ receptor inhibitor, and EGF, and in a condition in which cAMP is fed to the cells.
(FR) L’invention a pour objet de fournir un nouveau procédé permettant de fabriquer facilement une cellule présentant des fonctions proches de celles d’une cellule épithéliale intestinale du corps biologique, et une application de ce procédé. Selon ce procédé, une différenciation de la cellule souche pluripotente artificielle est induite dans la cellule épithéliale intestinale par : (1) une étape au cours de laquelle la cellule souche pluripotente artificielle est différenciée en cellule type endoderme ; (2) une étape au cours de laquelle la cellule type endoderme ainsi obtenue à l’étape (1) est différenciée en cellule type cellule souche intestinale ; et (3) une étape au cours de laquelle la cellule type cellule souche intestinale ainsi obtenue à l’étape (2) est différenciée en cellule type cellule épithéliale intestinale, et qui inclut une culture en présence d’un inhibiteur de MEK1, d’un inhibiteur de méthylation de l'ADN, d’un inhibiteur de récepteur TGF-bêta et d’un EGF, et dans des conditions d’alimentation des cellules en AMPc.
(JA) 生体の腸管上皮細胞に近い機能を示す細胞を簡便に調製可能な新たな方法及びその用途を提供することを課題とする。(1)人工多能性幹細胞を内胚葉様細胞へと分化させる工程と、(2)工程(1)で得られた内胚葉様細胞を腸管幹細胞様細胞へと分化させる工程と、(3)工程(2)で得られた腸管幹細胞様細胞を腸管上皮細胞様細胞へと分化させる工程であって、MEK1阻害剤、DNAメチル化阻害剤、TGFβ受容体阻害剤及びEGFの存在下、且つcAMPが細胞へ供給される条件下での培養を含む工程によって、人工多能性幹細胞を腸管上皮細胞へ分化誘導する。
