Abstract:
(EN) This invention relates to the field of small size pH-responsive tools, especially for measuring a pH value in biological media. I-motif DNA structures have been extensively used in the art for the fabrication of nanodevices; however, it is still a challenge to assist the folding of multi-stranded i-motif, especially at physiological pH. The inventors have conceived a multi- stranded i-motif-based pH-responsive tool, which comprises a poly-nucleic acid complex, instead of the mono-nucleic acid complexes which were known in the art. The poly-nucleic acid complex according to the invention is far easier to design, and may be easily adapted depending on the pH- range of interest to be measured. The poly-nucleic complexes of the invention are useful for designing a variety of pH-responsive devices, and especially pH biosensors and pH-sensitive delivery systems, particularly in the pharmaceutical field.
(FR) La présente invention concerne le domaine d'outils sensibles au pH de petite taille, en particulier pour mesurer une valeur de pH dans des milieux biologiques. Des structures d'ADN à motif en I ont été largement utilisées dans l'état de la technique pour la fabrication de nanodispositifs; cependant, l'assistance du repliement d'un motif en I multibrin, en particulier au pH physiologique, reste un défi. Les inventeurs ont conçu un outil sensible au pH basé sur un motif en I multibrin qui comprend un complexe de poly(acides nucléiques) au lieu des complexes de mono(acide nucléique) qui étaient connus dans l'état de la technique. Le complexe de poly(acides nucléiques) selon l'invention est nettement plus facile à concevoir et peut être facilement adapté en fonction de la plage de pH d'intérêt à mesurer. Les complexes polynucléiques de l'invention sont utiles pour la conception de divers dispositifs sensibles au pH et en particulier de biocapteurs de pH et de systèmes d'administration sensibles au pH, en particulier dans le domaine pharmaceutique.
