Abstract:
(EN) The invention is directed to a recombinant vanillyl alcohol oxidase protein (VAOr) which results from the fusion of VAO and MBP (maltose binding protein), wherein the VAO used is the vaoA gene of the fungus Penicillium simplicissimum. This recombinant protein is expressed in E. coli, is functional, has a high yield, a high enzymatic activity, is stable in a dimeric configuration, the histidine tail making it easier to purify the resultant enzyme. This enzyme can be used in natural processes for producing vanilla on a large scale.
(FR) L’invention concerne une protéine recombinante de vanillyl alcool oxydase (VAOr) qui est obtenue par fusion de VAO et de MBP (protéine de liaison au maltose, la VAO utilisée correspondant au gène vaoA du champignon Penicillium simplicissimum. Cette protéine recombinante s’exprime en E. coli, est fonctionnelle, avec un rendement élevé, une activité enzymatique élevée, stable sur le plan dimère, la queue d’histidine facilitant la purification de l’enzyme obtenue. Cette enzyme peut être utilisée dans des procédés de production naturelle de vanillyl à grande échelle.
(ES) La invención apunta a una proteína Vainillil alcohol oxidasa recombinante (VAOr) que resulta de la fusión de VAO y MBP (maltose binding protein, donde la VAO utilizada es el gen vaoA del hongo Penicillium simplicissimum. Esta proteína recombinante se expresa en E. coli, es funcional, con alto rendimiento, con alta actividad enzimática, estable en una conformación dimérica, donde la cola de histidina facilita la purificación de la enzima obtenida. Esta enzima puede emplearse en métodos de producción natural de vainilla a gran escala.
