Abstract:
(EN) The subject invention pertains to a Molecular Cell Diary System (MCDS), which allows identification of the history of somatic alterations in the cell. MCDS comprises one or more combinations of a DNA cutter and a DNA writer expressed under the control of a promoter controlled a cellular event of interest. The DNA cutter and the DNA writer are in a combination are co-expressed when an even of interest occurs. The DNA cutter creates double strand breaks (DSB) in a target DNA in a sequence specific manner and the DNA writer incorporates DNA sequences in the DSB. The endogenous DNA repair machinery synthesizes repairs the DSB. As such, the combination of the DNA cutter and the DNA writer modifies the target DNA and leaves "marks" of the occurrence of the cellular event of interest. These marks are sequenced and the cellular event history of the cell is deciphered.
(FR) L'invention concerne un système d'agenda cellulaire moléculaire (« Molecular Cell Diary System » ou MCDS) qui permet d'identifier l'historique d'altérations somatiques dans la cellule. Le MCDS comprend une ou plusieurs combinaisons d'un ciseau à ADN et d'une molécule d'écriture d'ADN exprimés sous le contrôle d'un promoteur contrôlé par un événement cellulaire d'intérêt. Le ciseau à ADN et la molécule d'écriture d'ADN sont combinés et sont coexprimés quand un événement d'intérêt se produit. Le ciseau à ADN crée des cassures double brin (« double strand breaks » ou DSB) spécifiques à la séquence dans un ADN cible et la molécule d'écriture d'ADN incorpore des séquences d'ADN dans la DSB. Le mécanisme de réparation d'ADN endogène synthétise et répare les DSB. Ainsi, la combinaison du ciseau à ADN et de la molécule d'écriture d'ADN modifie l'ADN cible et laisse des «traces» de l'occurrence de l'événement cellulaire d'intérêt.
