Abstract:
(EN) An enzymatic purification method involves the introduction of a sample comprising a target analyte and amino acids into a porous matrix of a reaction chamber. The reaction chamber includes first pores and second pores. The first pores contain polypeptide synthesis enzymes that react with the amino acids to form polypeptides. First pores having a first size to be accessible by amino acids but inaccessible by the subsequently formed polypeptides. The second pores have a second size greater than the first size, are in contact with the first pores and form a series extending from within the reaction chamber to a waste chamber. The formed polypeptides are migrated through the series of second pores to the waste chamber. The target analyte of the sample is extracted from the reaction chamber.
(FR) La présente invention concerne un procédé de purification enzymatique qui met en œuvre l’introduction d’un échantillon comprenant un analyte cible et des acides aminés dans une matrice poreuse d’une chambre de réaction. La chambre de réaction comprend des premiers pores et des deuxièmes pores. Les premiers pores contiennent des enzymes de synthèse de polypeptide qui réagissent avec les acides aminés pour former des polypeptides. Les premiers pores ont une première taille pour être accessibles par des acides aminés mais inaccessibles par les polypeptides formés consécutivement. Les deuxièmes pores ont une deuxième taille supérieure à la première taille, sont en contact avec les premiers pores et forment une série s’étendant de l’intérieur de la chambre de réaction à une chambre de déchets. Les polypeptides formés sont amenés à migrer à travers la série de deuxièmes pores vers la chambre de déchets. L’analyte cible de l’échantillon est extrait de la chambre de réaction.
