Abstract:
(EN) It has become clear that if a peptide having a thiol group near an amino group, and having, at the N-terminal, an amino acid residue, which introduced a specific protective group into the thiol group and amino group, undergoes translation by initiation suppression, the likelihood of an amino acid translation reaction being initiated will increase. Moreover, cleaved peptide generation will be suppressed, improving the translation efficiency and purity. Also, it has become clear that it is possible to efficiently promote a cyclisation reaction in the peptide due to an amide bond. Based on these findings, a new method for producing a peptide and nucleic acid complex, the peptide containing numerous unnatural amino acids and having a site for cyclisation due to an amide bond, was found.
(FR) La présente invention a trait à un procédé de production d'un complexe de peptide et d'acides nucléiques non cyclique. Selon l'invention, il est devenu évident que si un peptide présentant un groupe diol à proximité d'un groupe amino, et présentant, sur une extrémité n terminale, un résidu d'acides aminés, ayant introduit un groupe protecteur spécifique dans le groupe diol et le groupe amino, subit une traduction par suppression d'initiation, la probabilité qu'une réaction de traduction d'acides aminés soit amorcée augmentera. De surcroît, la génération de peptide clivé sera supprimée, améliorant l'efficacité de traduction et la pureté. De plus, il est devenu évident qu'il est possible de favoriser efficacement une réaction de cyclisation dans le peptide en raison d'une liaison amide. Sur la base de ces résultats, un nouveau procédé de production d'un complexe de peptide et d'acides nucléiques a été découvert, le peptide comportant de nombreux acides aminés non naturels et présentant un site de cyclisation en raison de la liaison amide.
(JA) チオール基をアミノ基近傍に有し、そのチオール基とアミノ基に特定の保護基を導入したアミノ酸残基をN末端に有するペプチドを、イニシエーション・サプレッション法により翻訳すると、アミノ酸翻訳反応の開始確率が向上するうえ、切断ペプチドの生成が抑制され、翻訳効率と純度が向上することが明らかとなった。また、当該ペプチドにおいて、アミド結合による環化反応を効率的に進行させることが可能であることが明らかとなった。これらの知見に基づき、非天然アミノ酸を多複数含むペプチドであってアミド結合による環化部位を有するペプチドと核酸の複合体の新たな製造方法が見出された。
