Abstract:
(EN) In order to provide an ambrein production method capable of easily obtaining ambrein, provided is a tetraprenyl-β-curcumene cyclase that: has an amino acid substitution at the fourth amino acid residue of a DXDD motif, from aspartic acid to an amino acid other than aspartic acid; (a) has a QXXXGX(W/F) motif and a QXXXX(G/A)X(F/W/Y) motif on the N-terminal side, and a QXXXGX(F/W/Y) motif, a QXXXGXW motif, and a QXXXGX(F/W) motif on the C-terminal side, but does not have the QXXXGXW motif at a site farther than 170 amino acid residues on the C-terminal side, with respect to the DXDD motif; (b) has an identity of 40% or more with an amino acid sequence represented by SEQ ID NO: 1 or 13; and (c) exhibits ambrein generating activity using squalene as a substrate.
(FR) L'invention a pour objet de fournir un procédé de fabrication d'ambréine permettant d'obtenir facilement une ambréine. Le but de l'invention est atteint au moyen d'une cyclase tétraprényl-béta-curcumène de type mutant dans laquelle un acide aspartique consistant en un résidu d'acide aminé occupant la quatrième position d'un motif DXDD est substitué par un acide aminé autre que l'acide asparique, et qui (a) possède avec ledit motif DXDD pour référence, un motif QXXXGX(W/F) et un QXXXX(G/A)X(F/W/Y) côté terminaison N, et un motif QXXXGX(F/W/Y), un motif QXXXGXW et un motif QXXXGX(F/W) côté terminaison C, mais ne possède pas de motif QXXXGXW en une position éloignée au-delà de 170 résidus d'acide aminé côté terminaison C, (b) présente une similitude avec une séquence d'acides aminés représentée par la SEQ ID No. : 1 ou 13, supérieure ou égale à 40%, et (c) développe une activité de génération d'ambréine avec un squalène pour substrat.
(JA) 本発明の目的は、簡便にアンブレインを得ることができるアンブレインの製造方法を提供することにある。 前記課題は、DXDDモチーフの第4番目のアミノ酸残基であるアスパラギン酸がアスパラギン酸以外のアミノ酸に置換しており、(a)前記DXDDモチーフを基準として、N末端側にQXXXGX(W/F)モチーフ、及びQXXXX(G/A)X(F/W/Y)モチーフ、C末端側にQXXXGX(F/W/Y)モチーフ、QXXXGXWモチーフ、及びQXXXGX(F/W)モチーフを有し、そしてC末端側に170アミノ酸残基以上離れた位置にQXXXGXWモチーフを有さず、(b)配列番号1又は13で表されるアミノ酸配列との同一性が40%以上であり、そして(c)スクアレンを基質としてアンブレイン生成活性を示す、テトラプレニル-β-クルクメン環化酵素によって解決することができる。
