Abstract:
(EN) The present disclosure provides methods and compositions for enhancing the processivity of a polymerase in catalyzing template-dependent DNA synthesis in high concentrations, more than 100 mM, of salt. Also disclosed are methods and compositions for enhancing the assembly of polymerase-template complex compatible with active DNA synthesis in the presence of levels of nucleotides at 0.8 - 2.2 micormolar and at a high temperature between 35 and 45°C, such as temperatures at or near the melting temperature of the polymerase. The complexes of template and polymerase may be used in nanopore sequencing.
(FR) La présente invention concerne des procédés et des compositions permettant d'améliorer la processivité d'une polymérase à catalyser la synthèse d'ADN dépendante d'une matrice à des concentrations fortes en sel, à savoir plus de 100 mM. L'invention concerne également des procédés et des compositions permettant d'améliorer l'assemblage du complexe polymérase-matrice compatible avec la synthèse active d'ADN en présence de taux de nucléotides à 0,8 à 2,2 micromolaires et à une température élevée située entre 35 et 45 °C, par exemple la température de fusion de la polymérase ou la température proche de celle-ci. Les complexes de matrice et de polymérase peuvent être utilisés pour le séquençage de nanopores.
