Abstract:
(EN) Methods and compositions herein provide non-naturally occurring γ-butyrolactones (GBLs) in racemic mixtures that increase efficiency and effectiveness of screening for production of antibiotics, and enhance yields and express silent pathways. Non-naturally occurring GBLs were synthesized and found to stimulate antibiotic production in several different streptomycete strains. Antibiotic production by Streptomyces coelicolor was induced by a racemic mixture of non-cognate stereoisomers of VB-D, seven of which are non-naturally occurring. Further, novel A-factor-type GBL analogs stimulated antibiotic production in S. coelicolor. Synthesis in response to the treatment with the non-cognates GBL was observed for known compounds including undecylprodigiosin, desferrioxamine and streptorubin B, as was synthesis of a compound of unknown structure. A group of 37 additional microbial strains was screened by principal component analysis to determine optimal concentrations of each of a panel of four non-cognate synthetic GBLs for addition to cultures with optimal stimulation of secondary metabolites, and large scale fermentations were analyzed and product enhancement by the GBLs was observed.
(FR) La présente invention concerne des procédés et des compositions permettant d'obtenir des gamma-butyrolactones (GBL) non naturelles dans des mélanges racémiques, qui augmentent l'efficience et l'efficacité du criblage en vue de la production d'antibiotiques, produisent des rendements plus élevés et expriment des voies silencieuses. Des GBL non naturelles ont été synthétisées et il a été constaté qu'elles stimulent la production d'antibiotiques dans différentes souches de streptomycètes. La production d'antibiotiques par Streptomyces coelicolor a été induite par un mélange racémique de stéréoisomères non apparentés de VB-D, dont sept sont non naturels. En outre, de nouveaux analogues de GBL de type facteur A ont stimulé la production d'antibiotiques par S. coelicolor. Une synthèse en réponse au traitement par les GBL non apparentées a été observée pour des composés connus, parmi lesquels l'undécylprodigiosine, la desferrioxamine et la streptorubine B ; la synthèse d'un composé de structure inconnue a également été observée. Un groupe de 37 souches microbiennes supplémentaires a été criblé par analyse en composantes principales pour déterminer des concentrations optimales de chacun d'un ensemble de quatre GBL synthétiques non apparentées en vue d'un ajout à des cultures avec une stimulation optimale de métabolites secondaires ; des fermentations à grande échelle ont été analysées, ce qui a permis de constater une augmentation de produit dû aux GBL.
