Abstract:
(EN) The invention relates to a system and method for introducing DNA into cells. In particular, the invention relates to a method for introducing single or multiple copies of a DNA sequence or gene of interest into a cell comprising providing: a) a "copy and paste" transposase; and b) a construct comprising a DNA sequence or gene of interest flanked by a "copy and paste" transposon terminal sequence, such as an LTS or RTS. A novel "copy and paste" transposon of the Helitron family is described along with systems for using the corresponding transposase in methods for introducing DNA into cells, for example, to generate cell lines for use in protein production, cell and gene therapy or as reference standards.
(FR) La présente invention concerne un système et un procédé d'introduction d'ADN dans des cellules. En particulier, l'invention concerne un procédé d'introduction de copies uniques ou multiples d'une séquence d'ADN ou d'un gène d'intérêt dans une cellule comprenant la préparation : a) d'une transposase "copier-coller"; et b) d'un produit de synthèse comprenant une séquence d'ADN ou un gène d'intérêt flanquée par une séquence terminale transposon "copier-coller", telle qu'une LTS ou RTS. Un nouveau transposon "copier-coller" de la famille Helitron est décrit, ainsi que des systèmes d'utilisation de la transposase correspondante dans des procédés d'introduction d'ADN dans les cellules, par exemple, pour générer des lignées cellulaires destinées à être utilisées dans la production de protéine, dans la thérapie cellulaire et génique ou comme étalons de référence.
