Abstract:
(EN) The present invention addresses the problem of providing a cultured transgenic cell and a transgenic animal, in which mouse norovirus (MNV) can grow beyond the barrier of host characteristics in mammalian cells, and use of the same in a screening method to thereby dissolve stagnation of studies on noroviruses. The present inventor has solved the aforesaid problem by finding that MNV infection is observed in a cultured transgenic mammalian cell, in which the whole or a part of mouse CD300F gene and/or CD300 family gene(s) being similar to mouse CD300F gene in a base sequence in an extracellular domain are incorporated, or a mammal having the aforesaid transgenic cells as own cells and, on the basis of this finding, providing a method for screening a norovirus-related drug etc.
(FR) La présente invention aborde le problème de la fourniture d'une cellule transgénique en culture d'un animal transgénique, dans lesquels un norovirus de souris (MNV) peut croître au-delà de la barrière des caractéristiques d'hôte dans des cellules de mammifère, et l'utilisation de ceux-ci dans un procédé de criblage afin d'éliminer la stagnation des études sur les norovirus. Le présent inventeur a résolu le problème ci-dessus en découvrant qu'une infection par MNV est observée dans une cellule de mammifère transgénique en culture, dans laquelle la totalité ou une partie du gène CD300F de souris et/ou un ou plusieurs gène(s) de la famille CD300 étant similaires au gène de CD300F de souris dans une séquence de bases dans un domaine extracellulaire sont incorporés, ou un mammifère comportant les cellules transgéniques mentionnées ci-dessus en tant que cellules propres et, sur la base de cette observation, propose un procéde de criblage d'un médicament lié à un norovirus, etc.
(JA) 本発明は、マウスノロウイルス(MNV)が、哺乳動物細胞内での宿主特性の壁を越えて増殖可能な遺伝子組換え培養細胞や遺伝子組み換え動物、さらにこれらを用いたスクリーニング方法の用途を提供して、ノロウイルスに関する研究停滞の問題を解決することを課題とする。本発明者は、マウスのCD300F遺伝子、及び/又は、当該遺伝子の細胞外領域の塩基配列が当該遺伝子に類似するCD300ファミリー遺伝子、の全部若しくは一部が一種又は二種以上組み込まれている、遺伝子組換え哺乳動物培養細胞、これを自己の細胞として保有する哺乳動物において、MNV感染が認められることを見出し、これを基にノロウイルスに関連する薬剤のスクリーニング方法等を提供することにより、上記課題の解決を行った。
