Abstract:
(DE) Die Erfindung betrifft ein Verfahren zur Ein-Topf-Ein-Schritt- Assemblierung von zwei oder mehr DNA-Molekülen zu mindestens einem rekombinanten DNA-Molekül sowie einen Träger und ein Kit hierfür. Aufgabe der vorliegenden Erfindung ist es, ein einfaches und kostengünstiges Assemblierungsverfahren für DNA-Moleküle bereit zu stellen. Zur Lösung der Aufgabe wird erfindungsgemäß ein Verfahren zur Ein-Topf-Ein-Schritt-Assemblierung von zwei oder mehr DNA-Molekülen zu mindestens einem rekombinanten DNA-Molekül bereit gestellt, wobei die zwei oder mehr zu assemblierenden DNA-Moleküle in trockener Form auf mindestens einem Träger vorliegend in einem Reaktionsgefäß mit einem geeigneten Reaktionsmedium zusammen gebracht werden.
(EN) The invention relates to a method for one-pot one-step assembly of two or more DNA molecules to form at least one recombinant DNA molecule, and a substrate and a kit for this purpose. The object of the invention is to provide a simple and cost-effective assembly method for DNA molecules. In order to achieve this object, a method for one-pot one-step assembly of two or more DNA molecules to form at least one recombinant DNA molecule is provided according to the invention, wherein the two or more DNA molecules to be assembled are brought together in dry form with a suitable reaction medium on at least one substrate present in a reaction vessel.
(FR) L'invention concerne un procédé pour réaliser l'assemblage de type « 1 tube-1 étape » de deux molécules d'ADN ou plus pour former au moins une molécule d'ADN recombinant ainsi qu'un substrat et un kit destinés à cet effet. L'invention vise à fournir un procédé simple et économique pour assembler des molécules d'ADN. À cet effet, l'invention propose un procédé pour réaliser l'assemblage de type « 1 tube-1 étape » de deux molécules d'ADN ou plus pour former au moins une molécule d'ADN recombinant, les deux molécules d'ADN ou plus à assembler étant mises en contact, sous forme anhydre, sur au moins un substrat présent dans un récipient réactionnel avec un milieu de réaction approprié.
