Acoustophoretic devices and methods for separating biological cells (particularly T-cells) from other fluids / materials using multi-dimensional acoustic standing waves are disclosed. The devices include an inlet, at least two outlets, and a flow chamber having an ultrasonic transducer-reflector pair. Specifically, T cells, B cells, or NK cells can be separated from other blood components. A dual-pass acoustophoretic system including two acoustophoretic devices arranged in series and fluidly connected to one another is also illustrated. Means for pre-chilling the mixture prior to separation in the devices or system can be used to improve retention, concentration, and clarification and to prevent outgassing.

La présente invention concerne des dispositifs et des procédés acoustophorétiques de séparation de cellules biologiques (particulièrement de cellules T) d’autres liquides/matériaux au moyen d'ondes stationnaires acoustiques multi-dimensionnelles. Les dispositifs comprennent un orifice d’entrée, au moins deux orifices de sortie, et une chambre d’écoulement présentant une paire de transducteur-réflecteur ultrasonores. Spécifiquement, les cellules T, les cellules B, ou les cellules NK peuvent être séparées d'autres constituants sanguins. Un système acoustophorétique à double passage comprenant deux dispositifs acoustophorétiques disposés en série et raccordés de manière fluidique l’un à l’autre est également illustré. La présente invention décrit un moyen de pré-refroidissement du mélange avant la séparation dans les dispositifs ou système afin d’améliorer la rétention, la concentration, et la clarification et d’empêcher le dégazage.
