Abstract:
(EN) Among conventional methods for differentiating pluripotent stem cells into desired cell types, a highly efficient differentiation induction method using human ES/iPS cells had yet to be established. Many attempts at such have been made, such as controlling the culture conditions and induction methods for stepwise differentiation through the addition of various cell proliferation factors and differentiation factors to the culture solution; however, the use of a complicated cultivation step is a major challenge. The present invention was arrived at by developing a method for reducing the undifferentiated state of pluripotent stem cells and inducing highly efficient differentiation into a desired cell type in a short period of time using pluripotent stem cells that readily undergo cell differentiation.
(FR) Parmi les procédés classiques de différenciation de cellules souches pluripotentes en types cellulaires souhaités, un procédé très efficace d'induction de la différenciation à l'aide de cellules ES/iPS humaines doit encore être établi. De nombreux efforts ont été réalisés, tels que le contrôle des conditions de culture et des procédés d'induction destinés à la différenciation par étape par l'ajout de divers facteurs de prolifération cellulaire et de facteurs de différenciation à la solution de culture ; cependant, l'utilisation d'une étape de culture compliquée est un défi majeur. La présente invention a été obtenue par le développement d'un procédé de réduction de l'état non différencié de cellules souches pluripotentes et d'induction de différenciation très efficace en un type cellulaire souhaité en un temps court à l'aide de cellules souches pluripotentes qui subissent facilement une différenciation cellulaire.
(JA) 従来の多能性幹細胞を所望の細胞型へ分化する方法では、ヒトES/iPS細胞を用いて高効率な分化誘導法は未確立であった。培養条件のコントロールや様々な細胞増殖因子・分化因子などを培養液に加えることによる段階的分化誘導法など多くの試みがなされてきたが、複雑な培養ステップの使用が大きな課題である。 多能性幹細胞の未分化性を低減させ、積極的に細胞分化する多能性幹細胞を用いて、短時間・高効率に所望の細胞型への分化誘導を行う方法を開発し、本発明を完成した。
