Abstract:
(EN) The invention is a method for fluorescence analysis of a sample in the course of which a fluorescence image is recorded and a fluorescent object identification step is carried out on the fluorescence image. The invention is, furthermore, is an apparatus for fluorescence analysis of a sample, the apparatus comprising a sample holder spatial region (10), an excitation light sources (12a, 12b), a fluorescence detector (14) adapted for recording a fluorescence image by imaging the fluorescent emitted light emitted under the effect of the fluorescent excitation light of a specific analysed sample volume present in the sample holder spatial region (10) during an analysis, and a holographic arrangement adapted for recording holographic images of the specific analysed sample volume, wherein the holographic arrangement comprising a holographic light source (16) and a holographic detector (18). In the apparatus the fluorescence detector (14) and the holographic detector (18) are arranged on a common optical axis with the sample holder spatial region (10) at opposite sides of the sample holder spatial region (10), and the apparatus comprises a coupling-in element (20) adapted for imaging an object wave originating from the holographic light source (16) onto the common optical axis and directing it towards the sample holder spatial region (10), and also adapted for reflecting the light of the holographic light source (16) and transmitting the fluorescent emitted light.
(FR) L'invention concerne un procédé d'analyse de fluorescence d'un échantillon, au cours duquel une image de fluorescence est enregistrée et une étape d'identification d'objet fluorescent est exécutée sur l'image de fluorescence. L'invention concerne en outre un appareil pour l'analyse de fluorescence d'un échantillon, l'appareil comprenant une région spatiale de porte-échantillon (10), des sources de lumière d'excitation (12a, 12b), un détecteur de fluorescence (14) conçu pour enregistrer une image de fluorescence par imagerie de la lumière émise fluorescente émise sous l'effet de la lumière d'excitation fluorescente d'un volume d'échantillon analysé spécifique et présent dans la région spatiale de porte-échantillon (10) pendant une analyse, et un agencement holographique conçu pour enregistrer des images holographiques du volume d'échantillon analysé spécifique, l'agencement holographique comprenant une source de lumière holographique (16) et un détecteur holographique (18). Dans l'appareil, le détecteur de fluorescence (14) et le détecteur holographique (18) sont disposés sur un axe optique commun avec la région spatiale de porte-échantillon (10) sur des côtés opposés de la région spatiale de porte-échantillon (10), et l'appareil comprend un élément de couplage (20) conçu pour imager une onde d'objet provenant de la source de lumière holographique (16) sur l'axe optique commun et pour la diriger vers la région spatiale de porte-échantillon (10), et conçu également pour réfléchir la lumière de la source de lumière holographique (16) et transmettre la lumière émise fluorescente.
