Abstract:
(EN) The invention relates to the field of passive immunotherapy of oncological diseases, and particularly to a method of adoptive cell transfer that uses genetically-modified autologous or allogeneic cytotoxic T-lymphocytes having acquired immunogenicity against tumour cells. The invention can be used in T-cell cancer therapy. The result is an original gene construct that encodes monomolecular chimeric T-cell receptors in which an effector fragment of a T-cell receptor is combined with an antigen-recognising portion in the form of variable fragments of two different carcinoembryonic antigen (CEA) antibodies. The cytotoxic activity toward CEA-expressing cells exhibited by human peripheral blood lymphocytes transfected by the described structures is higher than 50% of target cells.
(FR) L'invention concerne l'immunothérapie passive des maladies oncologiques, notamment une méthode de transfert adoptif de cellules utilisant des lymphocytes T cytotoxiques autologues ou allogènes génétiquement modifiés possédant une immunogénicité acquise vis-à-vis des cellules tumorales. L'invention peut s'utiliser dans la thérapie du cancer par lymphocytes T. On a créé une construction génétique originale codant pour les récepteurs chimères monomoléculaires de lymphocytes T dans lesquels un fragment effecteur du récepteur de lymphocytes T est regroupé avec une partie reconnaissant les antigènes qui se présente comme des fragments variables de deux anticorps différents vis-à-vis de l'antigène carcinoembryonnaire (CEA). La cytotoxicité des lymphocytes du sang périphérique humain transfectés par les constructions décrites ici vis-à-vis des cellules exprimant le CEA correspond à plus de 50 % des cellules cibles.
(RU) Изобретение относится к области пассивной иммунотерапии онкологических заболеваний, в частности к методу адоптивного переноса клеток, в котором используют генетически модифицированные аутологичные или аллогенные цитотоксические Т-лимфоциты, обладающие приобретенной иммуногенностью против клеток опухоли. И может быть использовано в Т-клеточной терапии рака. Создается оригинальная генетическая конструкция, кодирующая химерные мономолекулярные Т-клеточные рецепторы, в которых эффекторный фрагмент Т-клеточного рецептора объединен с антиген-распознающей частью, представляющей собой вариабельные фрагменты двух различных антител к раково-эмбриональному антигену (РЭА). При этом цитотоксическая активность трансфицированных описанными конструкциями лимфоцитов периферической крови человека в отношении РЭА-экспрессирующих клеток составляет более чем 50% клеток мишеней.
