Abstract:
(EN) Methods to qualitatively and quantitatively determine mRNA capping, and to determine 5' capping efficiency and 5' cap identity in RNA samples, all without the need for radiolabels, by using tagged probes that are complementary to the 5' end of target RNA and RNAse H to cleave the 5' end of RNA, then using LC-MS to determine the 5' RNA products.
(FR) L'invention concerne des méthodes pour mesurer qualitativement et quantitativement le coiffage de l'ARNm, et pour mesurer l'efficacité de l'addition de la coiffe en 5' et connaître l'identité de la coiffe en 5' dans des échantillons d'ARN sans devoir utiliser de radiomarqueurs, en utilisant des sondes marquées complémentaires de l'extrémité 5' de l'ARN cible et la ribonucléase H pour cliver l'extrémité 5' de l'ARN, utilisant ensuite la chromatographie en phase liquide couplée à la spectrométrie de masse (LC-MS) pour identifier les produits de l'ARN 5'.
