Abstract:
(EN) Herein is reported a method for determining the binding affinity of the binding sites of a bivalent full length antibody of the human IgG1 subclass to a homo-multimeric antigen comprising the steps of i) incubating a mixture comprising the antibody and a polypeptide that is derived from lysine-gingipain of porphyromonas gingivalis at a pH of from pH 7.5 to pH 8.5, in the presence of a reducing agent, at a temperature of from 30 °C to 42°C, for time of from 10 min. to 240 min. to cleave the antibody into Fabs and Fc-region, and ii) determining the binding affinity of the Fabs of the antibody for its antigen using a surface plasmon resonance method by directly applying the incubated reaction mixture obtained in the previous step in the surface plasmon resonance method and therewith determining the binding affinity of the binding sites of the bivalent full length antibody of the human IgG1 subclass.
(FR) L'invention concerne un procédé de détermination de l'affinité de liaison des sites de liaison d'un anticorps bivalent pleine longueur de la sous-classe IgG1 humaine pour un antigène homo-multimère comprenant les étapes consistant i) à incuber un mélange comprenant l'anticorps et un polypeptide dérivé de la lysine-gingipaïne de Porphyromonas gingivalis à un pH compris entre 7,5 et 8,5, en présence d'un agent réducteur, à une température de 30 à 42 °C, pendant une durée de 10 minutes à 240 min pour cliver l'anticorps en fragments Fab et Fc et ii) à déterminer l'affinité de liaison des fragments Fab de l'anticorps pour son antigène par un procédé de résonance plasmonique de surface par utilisation directe du mélange réactionnel incubé obtenu à l'étape précédente dans le procédé de résonance plasmonique de surface, ce qui permet de déterminer l'affinité de liaison des sites de liaison de l'anticorps bivalent pleine longueur de la sous-classe IgG1 humaine.
