Abstract:
(EN) Provided is a method for separating and extracting mesenchymal stem cells from the human umbilical cord. The method uses healthy neonatal umbilical cord tissue; after cleaning and disinfection, mechanically pulverising same, separating the Wharton's jelly, and after treating with erythrocyte lysate, carrying out suspension culture in a serum-free culture medium. Replacing the liquid every 3-5 days; after the plate adherence rate reaches 30-70%, carrying out trypsin digestion, and then collecting the cells by centrifugation for passage amplification, until the rate of confluence of the cells reaches over 90% confluence, thereby obtaining high purity umbilical cord mesenchymal stem cells.
(FR) L'invention concerne un procédé de séparation et d'extraction de cellules souches mésenchymateuses de cordon ombilical humain. Le procédé utilise un tissu de cordon ombilical néonatal sain et consiste à; après nettoyage et désinfection, pulvériser mécaniquement ledit tissu, séparer la gelée de Wharton et, après le traitement avec un lysat d'érythrocytes, effectuer une culture en suspension dans un milieu de culture sans sérum; remplacer le liquide tous les 3 à 5 jours; lorsque le taux d'adhérence de plaque atteint 30-70 %, effectuer une digestion à la trypsine, puis collecter les cellules par centrifugation pour une amplification par passage, jusqu'à ce que le taux de confluence des cellules atteigne plus de 90 % de confluence, ce qui permet d'obtenir des cellules souches mésenchymateuses de cordon ombilical de pureté élevée.
(ZH) 提供一种从人脐带分离提取间充质干细胞的方法。该方法利用健康新生儿的脐带组织,经过清洗消毒,机械剪碎,分离华通氏胶,经红细胞裂解液处理后以无血清培养基悬浮培养。每3-5天进行换液,平皿内贴壁率达30-70%后利用胰酶消化后,离心收集细胞传代扩增,至细胞汇合率达80-90%汇合,即得到高纯脐带间充质干细胞。
