Abstract:
(EN) Methods for preparing populations of hematopoietic stem cells (HSCs), e.g., autologous and/or allogenic HSCs, using mechanical stretching or transient receptor potential cation channel-subfamily vanilloid member 4 (Trpv4) agonists, and methods of use of the HSCs in transplantation are provided. Specifically, the methods comprising providing a population cells comprising hemogenic endothelial (HE) cells, contacting the HE cells with an amount of an agonist of Trpv4; and/or subjecting the cells to cyclic 2-dimensional stretching, for a time and under conditions sufficient to stimulating endothelial-to-HSC transition. Further disclosed are methods of treating malignant or non-malignant hematologic, metabolic, or immune disorders comprising administering to a subject a therapeutically effective amount of HSCs obtained by the methods.
(FR) L'invention concerne des procédés de préparation de populations de cellules souches hématopoïétiques (HSC), par exemple, des HSC autologues et/ou allogéniques, en utilisant un étirement mécanique ou des agonistes de Trvp4 (récepteur-canal cationique de potentiel transitoire, sous-famille vanilloïde, membre 4), et des procédés d'utilisation des HSC dans une transplantation. De manière spécifique, les procédés consistent à fournir une population de cellules comprenant des cellules endothéliales hémogéniques (HE), mettre en contact les cellules HE avec une quantité d'un agoniste de Trpv4 ; et/ou soumettre les cellules à un étirement bidimensionnel cyclique, pendant une durée et dans des conditions suffisantes pour stimuler une transition de cellule endothéliale à HSC. L'invention concerne aussi des procédés de traitement de troubles hématologiques, métaboliques ou immunitaires malins ou non malins consistant à administrer à un sujet une quantité à effet thérapeutique de HSC obtenues par les procédés.
