Abstract:
(EN) New methodologies for site-specifically radiolabeling proteins with the PET isotope [18F] are required to generate high quality radiotracers for imaging in both the preclinical and clinical settings. The enzymatic radiofluorination overcomes many of the limitations encountered to date with purely chemical approaches. The bacterial enzyme lipoic acid ligase was used to conjugate [18F]-fluorooctanoic acid to both a small peptide and a Fab antibody fragment. Labeling was site-specific and highly efficient under mild aqueous conditions using small amounts of peptide/protein (1-10 nmol). The labeled construct retained full epitope binding affinity and was stable in mouse serum. Using an optimized reaction scheme, mCi quantities of [18F]-Fab were generated, an amount sufficient for human imaging.
(FR) L'invention concerne de nouvelles méthodologies pour le radiomarquage spécifique de site de de protéines avec l'isotope de TEP [18F] qui sont nécessaires pour produire des indicateurs radioactifs de haute qualité pour l'imagerie en milieux précliniques et cliniques. La radiofluoration enzymatique surmonte de nombreuses limitations rencontrées jusqu'à présent avec des approches purement chimiques. L'enzyme bactérienne acide lipoïque ligase a été utilisée pour conjuguer l'acide [18F]-fluorooctanoïque à la fois à un petit peptide et à un fragment d'anticorps Fab. Le marquage est spécifique d'un site et très efficace dans des conditions aqueuses douces en utilisant de petites quantités de peptide/protéine (1-10 nmol). La construction marquée conserve toute l'affinité de liaison de l'épitope et est stable dans du sérum de souris. A l'aide d'un schéma de réaction optimisé, des quantités de l'ordre de la mCi de [18F]-Fab ont été produites, une quantité suffisante pour l'imagerie humaine.
