Abstract:
(EN) The present invention involves a ligand fluorescent sensor protein that specifically responds to ligands, leading to a change in fluorescence properties. The ligand fluorescent sensor protein includes a first fluorescent protein domain, an N terminal-side linker, a ligand-bonding domain, a C terminal-side linker, and a second fluorescent protein domain. The fluorescent protein used in the ligand fluorescent sensor protein has a β-barrel structure. The first fluorescent protein domain includes a β1–β3 β-sheet region from the N-terminal of the fluorescent protein, an α-helix region continuing therefrom, and a β4–β6 β-sheet region. The second fluorescent protein domain includes a β7–β11 β-sheet region of the same fluorescent protein as the first fluorescent protein domain. The N-terminal-side linker and the C-terminal-side linker are each independently a polypeptide comprising one or a plurality of amino acids.
(FR) La présente invention concerne une protéine capteur fluorescent de ligand qui répond spécifiquement à des ligands, entraînant un changement dans les propriétés de fluorescence. La protéine capteur fluorescent de ligand comprend un premier domaine de protéine fluorescente, un lieur côté N terminal, un domaine de liaison de ligand, un lieur côté C terminal, et un second domaine de protéine fluorescente. La protéine fluorescente utilisée dans la protéine capteur fluorescent de ligand présente une structure de tonneau β. Le premier domaine de protéine fluorescente comprend une région de feuillet β β1–β3 à partir de l'extrémité N-terminale de la protéine fluorescente, une région d'hélice α continuant à partir de cette dernière, et une région de feuillet β β4–β6. Le second domaine de protéine fluorescente comprend une région de feuillet β β7–β11 de la même protéine fluorescente que le premier domaine de protéine fluorescente. Le lieur côté N-terminal et le lieur côté C-terminal sont chacun, indépendamment, un polypeptide comprenant un ou plusieurs acides aminés.
