Abstract:
(EN) The present disclosure provides a method of identifying a substrate for a protease including contacting a protease to one of a first array and a second array, each array having the same plurality of features, each feature including at least one sequence linked to a solid support. The at least one sequence includes a candidate protease substrate linked to a reporter. The method further includes contacting a detectable element to each of the arrays to allow binding of the detectable element to the reporter, and detecting first and second signals resulting from binding of the detectable element to the each of the reporters in the first and second arrays. The method further includes comparing the first signal and the second signal to identify a difference in the first signal and the second signal, and identifying at least one candidate protease substrate as a substrate for the protease.
(FR) La présente invention concerne un procédé d'identification d'un substrat destiné à une protéase consistant à mettre en contact une protéase avec un d'un premier réseau et d'un second réseau, chaque réseau comportant la même pluralité de caractéristiques, chacun comprenant au moins une séquence reliée à un support solide. Ladite séquence comprend un substrat de protéase candidat relié à un rapporteur. Le procédé consiste en outre à mettre en contact un élément détectable avec chacun des réseaux pour permettre la liaison de l'élément détectable au rapporteur, et à détecter les premier et second signaux résultant de la liaison de l'élément détectable à chacun des rapporteurs dans les premier et second réseaux. Le procédé consiste en outre à comparer le premier signal et le second signal pour identifier une différence entre le premier signal et le second signal, et à identifier au moins un substrat de protéase candidat en tant que substrat destiné à la protéase.
