Abstract:
(EN) The present invention relates to a method for optimizing production of solid spawn using Saccharomyces cerevisiae Y204 (KACC93237P). Particularly, as a result of checking the optimal condition for producing solid spawn of Saccharomyces cerevisiae Y204 (KACC93237P), the following findings were confirmed: the growth rate and the number of live bacteria were the highest when a S. cerevisiae Y204 spawn was cultivated on a YPD culture medium for 24 hours at 25°C with a mixing rate of 150 rpm; spawn survival rate increased when 10% of skim milk, 1% of lactose, and 7 to 10% of trehalose were added, as excipients, when producing the solid spawn; freeze-drying showed 1.2 to 1.4 times fewer live bacteria but resulted in a contamination rate that was approximately twice as low compared to drying with hot air, when drying the solid spawn of S. cerevisiae Y204 enzymes. Therefore, the optimal condition for the solid spawn according to the present invention could be useful in the method for producing the solid spawn of Saccharomyces cerevisiae Y204 (KACC93237P).
(FR) La présente invention concerne un procédé d'optimisation de la production de descendance solide utilisant Saccharomyces cerevisiae Y204 (KACC93237P). Particulièrement, en résultat de la vérification des conditions optimales de production de descendance solide de Saccharomyces cerevisiae Y204 (KACC93237P), les découvertes suivantes ont été confirmées : la vitesse de croissance et le nombre de bactéries vivantes étaient les plus élevés lorsqu'une descendance de S. cerevisiae Y204 était cultivée sur un milieu de culture YPD durant 24 heures à 25°C sous une vitesse de mélange de 150 tr/min; le taux de survie de la descendance augmentait lorsque 10 % de lait écrémé, 1 % de lactose, et 7 à 10 % de tréhalose étaient ajoutés, comme excipients, lors de la production de descendance solide; la lyophilisation présentait 1,2 à 1,4 fois moins de bactéries vivantes mais résultait en un taux de contamination qui était approximativement deux fois aussi faible comparé au séchage à l'air chaud, lors du séchage de la descendance solide d'enzymes S. cerevisiae Y204. Par conséquent, les conditions optimales pour la descendance solide selon la présente invention pourraient être utiles dans le procédé de production de descendance solide de Saccharomyces cerevisiae Y204 (KACC93237P).
(KO) 본 발명은 사카로마이세스 세르비지애 (Saccharomyces cerevisiae) Y204(KACC93237P)을 이용한 고체종균 제조의 최적화 방법에 관한 것으로,구체적으로 사카로마이세스 세르비지애 (Saccharomyces cerevisiae)Y204(KACC93237P)의 고체종균을 제조하기 위해 최적조건을 확인한 결과, S. cerevisiae Y204 종균을 YPD 배지에서 교반속도 150 rpm으로 25°C 온도에서 24시간 배양하는 것이 생육도 및 생균수가 가장 높았고, 고체종균 제조 시 부형제로 skim milk 10%, lactose 1% 및 trehalose 7 내지 10%를 각각 첨가할 경우 종균 생존율이 높아짐을 확인하였으며, S. cerevisiae Y204 효모 고체종균 건조 시 동결건조가 열풍건조보다 1.2 내지 1.4배 정도 낮은 생균수를 나타냈으나, 2배 정도의 오염도가 낮음을 확인함으로써, 본 발명의 고체종균 최적화 조건은 사카로마이세스 세르비지애 Y204(KACC93237P) 고체종균 제조방법으로 유용하게 이용될 수 있다.
