Abstract:
(EN) Provided is a method of observing a living biological sample in an aqueous solution in a scanning electron microscope, with which observation can be performed with a higher resolution, and with which a composition analysis of the biological sample can be provided. The invention provides an observation method for observing an organic specimen such as an organism in an aqueous solution in a scanning electron microscope. In this method, an aqueous solution and an organic specimen are interposed between opposing surfaces of first and second insulating thin films forming an opposing pair, a pulsed electron beam the intensity of which varies in a pulse shape is scanned and radiated onto an electrically conductive thin film provided on an outward facing surface of the first insulating thin film, and an image is obtained corresponding to variations in the electric potential on an outward facing surface of the second insulating thin film, characterized in that a composition analysis of the sample is provided on the basis of the difference between images corresponding to pulsed electron beams having different ON/OFF frequencies.
(FR) L'invention concerne une méthode d'observation d'un échantillon biologique vivant dans une solution aqueuse au moyen d'un microscope électronique à balayage, permettant d'effectuer une observation à plus haute résolution et d'obtenir une analyse de composition de l'échantillon biologique. L'invention concerne une méthode d'observation permettant d'observer un spécimen organique, tel qu'un organisme dans une solution aqueuse, au moyen d'un microscope électronique à balayage. Selon la présente méthode, une solution aqueuse et un spécimen organique sont interposés entre des surfaces opposées de premier et second films minces isolants formant une paire en opposition, un faisceau électronique pulsé dont l'intensité varie selon une forme d'impulsion est balayé et rayonné sur un film mince électroconducteur disposé sur une surface orientée vers l'extérieur du premier film mince isolant, et une image est obtenue en correspondance avec des variations du potentiel électrique sur une surface orientée vers l'extérieur du second film mince isolant, caractérisé en ce qu'une analyse de composition de l'échantillon est obtenue sur la base de la différence entre des images correspondant à des faisceaux électroniques pulsés ayant des fréquences d'activation/désactivation (ON/OFF) différentes.
(JA) 走査電子顕微鏡内において水溶液中の生物試料を生きたままより高い分解能で観察できしかもその組成分析を与えるような観察方法の提供。 走査電子顕微鏡内で水溶液中の生物の如き有機物試料を観察する観察方法である。一対の対向する第1及び第2の絶縁性薄膜の対向面の間に水溶液とともに有機物試料を介在させ、第1の絶縁性薄膜の外向面上に与えられた導電性薄膜にその強度をパルス状に変化させたパルス電子線を走査照射し、第2の絶縁性薄膜の外向面上の電位変化に応じた画像を得る方法において、異なるON/OFF周波数のパルス電子線に対応した画像の差から試料の組成分析を与えることを特徴とする。
